UREA HYDROLYSIS TEST

Aim- To detect ability of given micro-organism to produce urease enzyme.                                 

    



Theory-  This test is useful to detect the ability of bacteria to produce urease enzyme. The enzyme is extra cellular induced enzyme. It is specifically act on urea as follows.





 
                                                                                                           Due to hydrolysis basic product is ammonia is produced because of ammonia pH is shifted from acidic to alkaline. In alkaline pH phenol red indicator turns to magenta colour. Then there is urease production indicates positive test.                                                       The urease enzyme is produced by organism Proteus vulgaris, Micrococcus spp etc. 

Diagram-

Composition -
. peptone- 0.5
. Yeast extract- 0.3
. Dextrose- 0.1
. L-Lysine- 0.5
. Bromocresol purple- 19ml
. PH- 6.8
. D/w- 100ml
                                  
Requirements-    
1. Sterile chirstensons's agar medium tube-
2. Suspension of given test organism. 


Procedure-


1.under aseptic condition streak the 

suspension of given test organism in zig-zag

manner.

2. Incubate at 37°C for 24 hrs.

3.After incubation observe the slant for 

change in colour 

4.Record the result.


Result- 


 Before incubation the slant in organ colour and after incubation the slant is observed in magenta colour. 





Conclusion- 


The given organism has ability to produce urease enzyme that hydrolysis the urea.
