Gelatin Hydrolysis Test

  Aim- To detect ability of given organism to hydrolyse gelatin or produce gelatinase enzyme or not.  Theory-  Some bacteria have ability to produce extra cellular enzyme like gelatinase. The gelatinase destroy or decompose gelatin.  Reactions- This reaction occurs in two sequential steps.  1. Gelatinase hydrolysis gelatin into polypeptide.  2. That produced polypeptide further converted into amino acids.            That amino acids use by micro-organism for metabolic purpose gelatin hydrolysis test is use in classification and identification of organism. Hydrolysis of gelatin is detected by flooding the plate with Hgcl2. Requirements-  1. Gelatin agar plates.  2. 24hrs old culture of test organism (psuedomonas)  3. Hgcl2. Procedure-  1. Preparation of media (gelatin agar)

  Aim- To prepare gelatin agar medium laboratory.  Theory- Gelatin is protein derive from the animal protein collagen. It is component of connective tissue and tendons gelatin dissolve with water at 50 ℃ exists as liquid above 25℃. solidify as gets when cool below 25℃ is prepared by boiling of skin and bones gelatin agar medium. It is used as  biochemical test. This medium is to perform gelatin hydrolysis test. In this medium gelatin is rarely use as substrate because it is attached to enzyme and decompose by bacteria nutrient agar is enzyme and decompose by bacteria nutrient agar is use as basal medium KH 2 PO 4  and KH 2 PO 4  is use as buffer and glucose is acts as carbon source.
